<p><br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 男 1968年8月出生 博士 中共党员。1992年至今在广西医科大学口腔医院工作，教授，科室主任，硕士研究生导师。自1992年参加工作以来，一直从事口腔颌面外科的教学、科研以及临床工作。其中1994年至1997年在广西医科大学口腔系口腔颌面外科专业攻读硕士学位，研究课题是恶性肿瘤淋巴道转移化疗的研究。1997年至2000年在华西医科大学口腔医学院攻读博士学位，研究课题是淋巴管内皮细胞生长因子与恶性肿瘤淋巴道转移机理的研究</p>
<p>&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 在临床工作中，坚持为人民服务的宗旨，对病人一视同仁，多次拒收红包。工作认真负责，对待病人态度好，多次获得病人表扬，无医疗差错事故。在临床操作技能、诊断、治疗等方面有较深造诣。积极创新，对自己掌握的技术不保守，热心向下级医生传授自己的心得体会。擅长口腔颌面外科的各种手术操作。门诊治疗方面，擅长口腔内、外、修复科的综合治疗以及序列治疗。获得2003、2004年度优秀职工，优秀党员。</p>
<p>　　在教学工作中，无论是本科生、专科生、研究生以及进修生，都能做到勤勤恳恳，认真备课，虚心向其他教师请教教学经验；钻研先进的教学措施，革新教学手段，与科室同事进行了专业教学的改革试验，获得学生好评。认真带教，讲课内容翔实，生动风趣，深得学生好评。2002年开始招收硕士研究生，目前在培研究生2人。2002年度被评为广西医科大学十大杰出青年教师。2002年获得国家留学基金资助，赴芬兰访问学者一年。2004年十佳青年学术研究标兵,卫生厅科研先进个人。</p>

            <div><span style="COLOR: black">1</span><span style="COLOR: black">、</span><span style="COLOR: black">1995</span><span style="COLOR: black">年在国际上首先开展了口腔恶性肿瘤淋巴化疗的研究，采用先进的控释、靶向淋巴药物载体，携带化疗药物，选择性的进入肿瘤引流区淋巴管及淋巴结缓释，达到杀灭淋巴转移灶的目的，在实验研究中取得了满意的效果。目前正在进行进一步的临床前研究，准备进行临床研究。</span>肿瘤淋巴道转移很多见，相当多的患者不是死于原发灶, 而是死于不同程度的转移, 出现转移, 患者的被治愈率和生存率明显降低。目前常规的全身化疗，极少到达淋巴系统，对淋巴转移效果极差。且付作用大，效率低下，大量的药物流失到机体其他部位，在治疗肿瘤的同时有强大的毒付作用；而手术损伤大，费用高；放疗损伤大，效果差。目前国内外淋巴化疗的研究不多，还没有有效的药物能进入临床。因此，发展一种新型的缓/控释、靶向淋巴系统化疗药物，对于提高治愈率、生存率和生存质量，使我国的肿瘤治疗达到世界先进水平；针对国情，简化治疗方法，降低治疗费用；向基层推广，以满足广大人民群众的健康需要；将产生十分明显的社会效益及经济效益。</div>
            <div><span style="COLOR: black">2、<span style="FONT: 7pt 'Times New Roman'">&nbsp;&nbsp;&nbsp; </span></span><span style="COLOR: black">1997</span><span style="COLOR: black">年在国内首先开展了</span><span style="COLOR: black">VEGF-C</span><span style="COLOR: black">的研究。在国家自然科学基金、广西自然科学基金的资助下，在口腔癌淋巴道转移、癌周淋巴管密度三者的关系研究上取得了重要的进展。在国内首先提出</span><span style="COLOR: black">VEGF-C</span><span style="COLOR: black">是口腔癌淋巴道转移的重要标志物，口腔癌</span><span style="COLOR: black">VEGF-C</span><span style="COLOR: black">的表达增高，与癌周淋巴管密度的增加、淋巴道转移的增加密切相关。</span>已完成的科研取得了满意的结果，并得到此后国内外学者相关实验结果的证实。<span style="COLOR: black">目前，正在进一步进行重组</span><span style="COLOR: black">VEGF-C</span><span style="COLOR: black">技术，相关的临床前研究，以及相关抗体、检测试剂盒的研究。</span></div>
            <div><span style="COLOR: black">3、<span style="FONT: 7pt 'Times New Roman'">&nbsp;&nbsp;&nbsp; </span></span><span style="COLOR: black">2000</span><span style="COLOR: black">年开始在国家自然科学基金的资助下，国内首先开展了</span>VEGF基因转染恢复放疗后组织血管生成的实验研究。采用基因工程方法，通过逆转录PCR法构建了VEGF穿梭质粒，并建立了成熟的基因检测，转染方法，并在肌肉组织表达VEGF蛋白。构建了稳定的小鼠放疗模型，获得稳定的放疗动物缺血肌肉转染方法，并用定量RT-PCR、免疫组织化学法分别检测phVEGF165基因和蛋白表达，进行转染前后的比较，以确定转染是否成功。进行转染前后的组织内血管生成解剖学、生理学参数比较，评价转染对缺血区血管生成的作用，探讨今后应用于临床的可能性。试验获得可喜结果，成果已经通过自治区科技厅和教育厅成果鉴定，荣获广西科技进步三等奖。</div>

<p><span style="FONT-SIZE: 10.5pt">4</span><span style="FONT-SIZE: 10.5pt">、</span><span style="FONT-SIZE: 10.5pt">多药耐药基因（</span><span style="FONT-SIZE: 10.5pt">mutidrug resistance,MDR</span><span style="FONT-SIZE: 10.5pt">）和多药耐药相关蛋白（</span><span style="FONT-SIZE: 10.5pt">mutidrug resistance-associated protein,MRP</span><span style="FONT-SIZE: 10.5pt">）基因耐药表型是头颈癌等肿瘤最重要的细胞防御机制。头颈癌是我国最常见的恶性肿瘤之一，口腔癌占全身恶性肿瘤的</span><span style="FONT-SIZE: 10.5pt">1.9%~3.5%,</span><span style="FONT-SIZE: 10.5pt">居头颈恶性肿瘤的第二位，仅次于鼻咽癌。目前，治疗仍然以综合治疗最为有效。其中放疗是治疗的重要手段，也是必不可少的。临床上发现有的患者对放疗不敏感，有的患者在放疗后对后续的治疗不敏感。因此严重影响了肿瘤的根治及增加了复发或转移的发生率。这种对放疗不敏感的主要原因在于癌细胞的内在性或获得性的耐药性，这种耐药性不仅对于放疗，而且对于化疗的不敏感也与之密切相关，不过由于有相同的分子机制，也称之为耐药性。</span><span style="FONT-SIZE: 10.5pt">2002</span><span style="FONT-SIZE: 10.5pt">开始，在学校博士启动基金的资助下，进行</span><span style="FONT-SIZE: 10.5pt; COLOR: black">多药耐药基因</span><span style="FONT-SIZE: 10.5pt; COLOR: black">MDR1/MRP</span><span style="FONT-SIZE: 10.5pt; COLOR: black">反义寡核苷酸逆转舌癌细胞放疗耐药性的研究。</span><span style="FONT-SIZE: 10.5pt">我们首先利用人舌癌细胞株</span><span style="FONT-SIZE: 10.5pt">Tca8113</span><span style="FONT-SIZE: 10.5pt">建立起一定剂量下的耐放疗细胞株，然后我们利用</span><span style="FONT-SIZE: 10.5pt">ANOS</span><span style="FONT-SIZE: 10.5pt">的作用特点，设计了特异针对</span><span style="FONT-SIZE: 10.5pt">MDR1</span><span style="FONT-SIZE: 10.5pt">和</span><span style="FONT-SIZE: 10.5pt">MRP</span><span style="FONT-SIZE: 10.5pt">基因的反义寡核苷酸，利用目前先进的脂质体</span><span style="FONT-SIZE: 10.5pt">Lipofectamine</span><span style="FONT-SIZE: 10.5pt">作为载体，转染我们建立的耐放疗细胞株，通过实验，达到了降低该耐放疗细胞株的耐药性的目的。这一方法的成功为其过渡到临床提供可靠的依据，并且为临床治疗创造一种新的基因治疗方法：结合目前放、化疗的新模式，同时为肿瘤的复发和转移的预防和治疗开辟一条崭新的途径。该研究培养的硕士研究论文获得了学校优秀硕士论文。</span></p>

<title>肿瘤及抗肿瘤药的概述</title>

<p>一、世界恶性肿瘤的现状<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 肿瘤是严重威胁人类生命健康的疾病，据世界卫生组织调查表明，全球肿瘤患者比例约占6.89&permil;，而每年新增肿瘤患者为1.78&permil;，生存数近4000万人。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 1996年全球58亿人口中，因癌症而死亡的达600余万人，占总死亡人数的近12%。另有新诊癌症患者1000余万人，估计癌症患者数4000余万人。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 在西方发达国家，常见肿瘤依次为：乳腺癌、肺癌、前列腺癌、结直肠癌、卵巢癌。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 随着科学的进步，以及检测手段和治疗技术的完善，使肿瘤得以早发现、早治疗，存活率不断提高。与此同时，总体存活率得到了增长。最新报道，美国2002年癌症的死亡率已呈现平稳下降的趋势。</p>
<p>二、我国恶性肿瘤的现状<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; （1）我国城乡地区居民肿瘤患病率分析&nbsp;&nbsp;&nbsp; <br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 近三十年来，我国癌症发病率持续上升，也有少数肿瘤的发生率较稳定或下降。1973年至1975年，我国因恶性肿瘤死亡的人数占全国总死亡人数的比例为12.6%，20世纪90年代为17.9%。国家卫生部统计资料表明，目前全国恶性肿瘤患病率达到1.15～1.27&permil;，良性肿瘤患病率达到0.93&permil;。发病率较高的恶性肿瘤的种类也有所变化。20世纪50年代至60年代，胃癌、食管癌、宫颈癌的发病率最高。近几年，宫颈癌、鼻咽癌及食管癌的发病率都有所下降，其中宫颈癌的发病率下降幅度最明显，下降了60%。目前，在大、中城市里，肺癌、乳腺癌发病率最高，在农村地区仍然是胃癌、食管癌的发病率最高。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; （2）我国肿瘤疾病谱的变化&nbsp;&nbsp;&nbsp; <br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 国家卫生部统计资料表明，我国恶性肿瘤全国患病率达到1.15&permil;，以全国总人口为基数，1998年我国恶性肿瘤患者约有148.5万左右，2001年我国恶性肿瘤患者约有145～160万人。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 我国常见肿瘤依次是胃癌、肝癌、肺癌和食管癌，其前五位占总体的74.34%，每年接受放、化疗患者数约为100多万人。&nbsp;&nbsp;&nbsp; <br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 从肿瘤死亡率看，目前在各种癌症中死亡率最高的是胃癌、肝癌、肺癌及食管癌。每年死于这四种恶性肿瘤的患者人数约占全部癌症患者死亡人数的70%左右。每年全国肿瘤死亡人数约有154万人左右，城市死亡率略高于农村死亡率。这与全国212.7万新增患者（新诊率）相比有近60万的落差，今后我国每年的现有肿瘤患者人群正以每年约60万人的速度膨胀。</p>
<p>三、我国抗肿瘤药市场分析<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; （1）概况<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 我国抗肿瘤药物的研究开发始于50年代末期，60年代初已有生产品种。目前我国抗肿瘤药物的开发已从无到有，从少品种而发展到系列化生产。30多年来，特别是近10年来，我国抗肿瘤药物的开发取得令人瞩目的成就。目前，我国抗肿瘤药物生产企业已发展到近百家，其中主要原料药厂20多家，制剂厂和中药厂有60多家。迄今为止，我国抗肿瘤药物已发展到7大类，160多个品种。其中烷化剂类有环磷酰胺、氮芥、噻替派等30多个品种；抗生素类有阿霉素（多柔比星）、丝裂霉素、争光霉素、正定霉素等20多种；植物类（天然类）药有长春新碱、秋水仙碱、喜树碱、紫杉醇、绿舒筋、复方醋酸棉酚等40多种；抗代谢类药有硫嘌呤唑、氟尿嘧啶、阿糖胞苷等20多种；杂类有丙脒腙、甲苄肼、门冬酰胺酶等13种以及激素类10多种和辅助类药近10种。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 目前国际上公认疗效较好的常用抗肿瘤药物有50-60种，我国大多数具有生产能力。WHO组织公布的常用抗肿瘤药物49个，我国能生产的就达40种以上，占86%以上。目前我国年生产抗肿瘤药物30多吨，主要分布在上海、江苏、浙江、山东等地。但远远不能满足临床日益增长的需要，还要依赖进口来弥补国内的不足，故开发生产抗肿瘤药物市场潜力巨大。在这几类常用抗癌药物中，最具特色的是植物类（天然类）药，特别是由于近年来国际上对该类药物研究开发的高潮不断，更加加速了该类用药水平的不断提高，像紫杉醇、喜树碱衍生物等新品种上市以来，由于确切的疗效和较小的毒副作用，越来越受到临床医生和肿瘤患者的青睐，该类用药在肿瘤大家族中已经确立了不可替代的地位。</p>
<p>&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; （2）我国抗肿瘤药物医院市场趋势分析&nbsp;&nbsp;&nbsp; <br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 近几年，我国抗肿瘤用药市场得到长足发展，用药市场规模不断扩大，2002年市场规模较1999年翻一番，达到70亿元人民币，每年的用药金额增长速度均在25%以上，其中2001年的增长速度高达50%以上。虽然2002年增长率较2001年明显下降，但预计今后几年仍有较好的发展。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; &nbsp; （3）我国抗肿瘤药物医院市场各类别用药状况&nbsp;&nbsp;&nbsp; <br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 近几年来，植物生物碱和其它天然药这一类别一直是抗肿瘤用药的领先类别，市场份额（金额百分比）从1999年的39.1%上升到2001年的39.23%，2002年继续稳步增长达43.99%。细胞毒素类抗生素和相关物质近年来的市场份额均在20%左右，从2001年的23.01%下降到2002年的18.05%，市场处于稳定阶段。</p>

<p>四、抗肿瘤药的未来<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 目前抗癌药物占全球药品市场总销售额的4.5%，所占份额虽小于其它大类药，据国外信息刊物公布的数字，世界抗肿瘤药物市场的年均增长率达12%以上。目前世界上抗肿瘤药物最畅销的主要品种除紫杉醇（目前销售额为10亿美元左右）外，还有吉西他宾（Ｇｅｍｃｉｔａｒｂｉｎ）、拓朴替康（Ｔａｐｏｔｅｃａｎ）和多西紫杉（Ｔａｘｏｔｅｒｅ）。后三种抗肿瘤药物的世界销售额合计超过4亿美元，三年后可望超过10亿美元。这三种药治疗发病率较高的肺癌、乳癌、卵巢癌、结肠癌、直肠癌和前列腺癌均有出色表现，目前尚无更好的其他药物可替代，是很有发展前途的产品。　<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 据国外报道，目前除已上市的单克隆抗体、肿瘤坏死因子、干扰素、白介素等生物工程药品（合抗癌疫苗）外，西方国家正在开发研制中的抗肿瘤药物有以下8大类：<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ①抗代谢药如二氢叶酸还原酶抑制剂、ＴＳ抑制剂、多位点抗叶酸代谢药、ＧＡＲＴ和ＡＶＣＡＲＴ抑制剂等。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ②作用于细胞骨架的抗肿瘤药物如紫杉醇（可抑制微管蛋白的合成）和作用于微丝合成的新型抗癌药（主要来自海洋生物体内，可利用生物工程技术来生产）。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ③法尼基转移酶抑制剂。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ④拓朴异构酶抑制剂如喜树碱及其系列衍生物依托泊甙、柔红霉素等。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ⑤还原型谷胱甘肽（ＧＳＨ）调节剂。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ⑥肿瘤细胞表面受体功能抑制剂。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ⑦核苷转运抑制剂如来自绿茶的&ldquo;茶多酚&rdquo;。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; ⑧基质金属蛋白酶抑制剂。　　<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 抗癌药物市场一般是指销售用于治疗5种主要类型的癌症（乳腺癌，结肠直肠癌，肺癌，卵巢癌和前列腺癌）的药物。在7个主要的医药市场中，大多数癌症的发生率正在升高。然而，5个主要的治疗癌症的药物增长是受人口转换的驱动，而不是该疾病发展危险的增加。乳腺癌目前是最流行的癌症，其后分别为前列腺癌，结肠直肠癌，肺癌和卵巢癌。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 在2000-2005年期间，化合物上市的最大机会是在胰腺癌和肺癌的适应症上。据决策资源公司估计，血管生成抑制剂市场在2004年和2010年将分别达到5.3亿和64亿美元。基质金属酶抑制剂和信号转导抑制剂市场2004年将达27亿美元。2010年整个抗癌药物市场将超过120亿美元。用于癌症治疗的单克隆抗体治疗药在今后几年将在世界七个主要市场具有惊人的增长。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 报告指出，预期在这些市场的销售额将从1998年的4.99亿美元增至2008年的44亿美元。这些产品现在的主要靶向为乳腺癌、卵巢癌、非小细胞肺癌、直结肠癌急性白血病和非何杰金氏淋巴癌。另外，ＤＲ公司也提出了一份对这些市场的急性白血病治疗药物前景的研究报告，预计到2008年全球急性白血病治疗药物总销售额将接近15亿美元，年增长率为16%。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 抗肿瘤用植物药是抗肿瘤用药的主体，占41%的市场份额，这与这类药的毒副作用相对较小有关。从上世纪90年代上市的抗肿瘤药中，除米托葸醌、羟基脲和顺铂等少数几种抗肿瘤药为化学合成药外，已上市(或仍在研制阶段)的一系列抗肿瘤药(含人体免疫剂)均为天然草药中提取的成分。浙江康莱特药业有限公司生产的康莱特注射液（薏苡仁油）就是一个典型代表，己获美国FDA审评批准，作为处方药在美国新药临床试验医院开始为期4个月的人体临床试验。今后几年中国市场上很可能涌现出一大批类似的新型天然类抗肿瘤药物。现已证实中药抗肿瘤活性成分有莪术油、华蟾素、黄芪多糖、冬虫夏草多糖、绿茶素、苦参碱、紫苏油、云芝多糖、枸杞多糖、大蒜素等。预计天然抗肿瘤药将成为今后抗肿瘤药物市场上的主角。 </p>
<p>　　除抗肿瘤用药以外，抗肿瘤辅助用药也占有较大的市场份额，主要为免疫刺激剂和免疫抑制剂。在免疫刺激剂类抗肿瘤药物中，生物工程药品则占有很重要的地位，如重组粒细跑集落刺激因子和干扰素。随着人类基因组计划的完成和基因生物研究的迅速发展，人类对肿瘤发病机理及其相关生物特性的更多了解和把握、癌细胞更多、更稳定的特异性抗原的发现以及生物芯片等基因检测工具的推广应用所带来的个体特异化的抗肿瘤治疗方法，将使血管生成抑制剂、癌疫苗、基因治疗、新单克隆抗体药物成为以后肿瘤治疗的主要趋势。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 抗肿瘤用药市场将不断扩大。据相关报告显示，中国抗肿瘤用药市场目前仍呈不断扩大的趋势。据有关部门对上海、北京、广州、沈阳等14个城市200多家医院抗肿瘤用药金额的统计，1999年、2001年、2002年上述区域各医院抗肿瘤用药的金额均持续上升，平均上升幅度在20%以上。有关研究表明，1999年国内抗肿瘤药物市场规模为60.3亿元，2001年为70.2亿元，2002年为82.13亿元，市场规模增长明显。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 国家行业部门统计表明，近几年我国重点城市典型医院抗肿瘤用药金额呈上升趋势，2002年同比增长14.64%，已达15.22亿元，市场规模增长明显。据中国医药商业协会报道，2001年抗恶性肿瘤药品占3.35%，从不同角度分析，比上一年增长了17～20%。<br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 从国家食品药品监督管理局2002年到2003年上半年对全国十几个大城市的主要医药公司的调查来看，抗肿瘤药物销售量占总体药品销售量比例稳步上升，2002年第二季度所占比例为0.94%，2003年同季度上升为1.05%，同比增长11.7%；抗肿瘤药物销售额占总体医药市场比例虽然各季度起伏较大，但总体上仍呈上升趋势。预计2003年中国抗肿瘤药物的销售金额将达到10.19亿美元，而2004年该数字将上升至11.98亿美元，2005年全国抗肿瘤药物市场销售将达到16.96亿美元。&nbsp;&nbsp;&nbsp; <br />&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; 未来几年内，对发病率高的胃癌、肝癌、肺癌和食管癌等，其对症治疗药物以及新型抗癌药物将成为医药市场上的新热点。目前对急性白血病的治疗还没有理想的药物，临床对该病还束手无策，其用药有待进一步研究开发。<br /></p>]]>

<p class="MsoPlainText" style="MARGIN: 0cm 0cm 0pt -1.75pt; TEXT-INDENT: 28.5pt"><font face="宋体" size="3">我们首次用物理凝聚法成功地将常规化疗药制备成新型固脂毫微粒。固脂毫微粒具有含药量高，稳定性，生物相容性好及毒性低等优点，便于制备和灭菌，避免传统的各种药物载体存在药物从载体中渗漏或骨架没有足够稳定性的缺陷。在此基础上，通过均匀设计法，优化了固脂毫微粒的制备工艺，使其外观、再分散性及粒径等指标均优于传统胶体微粒制剂。</font></p>

<p>[摘要] 目的：检测新型缓释剂载体多孔玻璃微粒（Porous Glass Particles，PGP）携带化疗药物局部注射的缓释及靶向淋巴系统能力。方法 比较大鼠舌粘膜下注射吸附平阳霉素(PYM)的PGP水溶液（PYM－PGP），PYM水溶液（PYM－SOL），以及静脉注射PYM水溶液（PYM－SOL-iv）不同时间段舌注射局部及周围组织、颌下淋巴结、血浆PYM浓度。结果 在舌部和颌下淋巴结组织PYM－PGP组PYM浓度，持续时间明显高于PYM－SOL组，PYM－SOL-iv组，而血浆中PYM－PGP组PYM浓度，持续时间明显低于PYM－SOL组，PYM－SOL-iv组。结论 PGP吸附PYM局部注射做为一种口腔淋巴化疗的新手段，可望提高疗效，减少用药量和毒副作用，达到对原发灶和淋巴转移双重治疗的目的。 <br />关键词：多孔玻璃微粒 平阳霉素 淋巴化疗&nbsp;<br />&nbsp;</p>

<div>Objective: to investigate the sustain release and target to lymphatic system ability of new sustain release drug carrier, Porous Glass Particles(PGP) absorbing chemotherapic drug local&nbsp;injection．<span>Methods: the effect of submucous injection of PYM-PGP, PYM solution(PYM-SOL) in rat tongues and intravenously injection of PYM-SOL (PYM-SOL-iv) was compared. Results: both the PYM concentration and sustain time of the PYM-PGP group were greatly higher than the other two groups in injection site and in submandibular lymph nodes, but both remarkabley lower than the other two groups in blood plasma. Conclusion: PYM-PGP can be used as a new chemotherapy method simutanously to treat primary tumor and lymph metastasis, to reduce drug dosage and toxity and enhance efficacy of drug.</span></div>

In the therapeutic principle of above diseases, promotion of the blood and lymphatic vessels regeneration is very important. Until now there are still not effective non-operation methods, especially correlated drug. </font></span></p>
<p class="MsoNormal" style="MARGIN: 0cm 0cm 0pt"><span lang="EN-US"><font size="3"><span style="mso-spacerun: yes">&nbsp;&nbsp; </span>In <st1:country-region w:st="on"><st1:place w:st="on">China</st1:place></st1:country-region>, yearly Incidence of malignant tumor is about 0.12%. That means that there are approximately 1.6 million of malignant tumor patients. Most of these patients did not die of primary tumor, but of different tumor metastasis, especially lymph node metastasis which rate of all cases reaches above 30%. Once tumor metastasis appearance, the cured rate and survival rate will obviously decrease. Because earlier period metastasis is difficult to detect by pathologic examination, there may be more cases. Many cases lose the best opportunity to be cured when the metastases having been detected. The cure rate and survival rate obviously decrease.<span style="mso-spacerun: yes">&nbsp;&nbsp; </span></font></span></p>
<p class="MsoNormal" style="MARGIN: 0cm 0cm 0pt"><span lang="EN-US"><font size="3"><span style="mso-spacerun: yes">&nbsp;&nbsp; </span>VEGF-C (vascular endothelial growth factor-C) was discovered in 1996, which belongs to the PDGF/VEGF family. Its receptors are VEGFR-2 and VEGFR-3. VEGF-C regulates vascular permeability and angiogenesis through VEGFR-2 and lymphangiogenesis through VEGFR-3 which is mainly expressed in the endothelium of lymphatic vessels. In clinic, as a therapeutic angiogenesis method, VEGF-C gene and recombinated protein can be used as effective blood and lymphatic vessels regenerated agents to treat ischemic heart dieases, extremity ischemia and lymphoedema. The agents are efficient and good curative effect in experiment and clinic study. But clinic use of the drug is not reported at present.
